Reserving the room in 25Live is required, regardless of whether you are using Teams and have added the room to the location of the Teams meeting. The person that has the room reserved through 25Live will be entitled to the space when conflicts occur. 

To enable your Teams meeting to work with this room, please make sure to update the location of your meeting to include the room you have reserved.  For more information, click here.
